Erst ab einer Dicke von 0,03 bis 0,02 mm (30 bis 20 µm) ist eine Probe für die Durchlichtmikroskopie geeignet und eine eingehendere Untersuchung wird möglich. Untersucht wird im normalen und polarisierten Licht sowie mit speziellen Wellenlängen (z. B. UV-Licht).
